Exploitation of the pearl oyster Pinetada margaritifera, which has developed in French Polynesia over the last twenty years, is based mainly on the collection of natural spat. Recruitment is highly variable in space and time and so spat is very often transferred by the farmers between atolls or even between archipelagos. Pearl formation: Persistence of the graft during the entire process of biomineralization ArchiMer

Most bivalves species of the genus Pinctada are well known throughout the world for production of white or black pearls of high commercial value. For cultured pearl production, a mantle allograft from a donor is implanted into the gonad of a recipient oyster, together with a small inorganic bead. Because of the dedifferentiation of cells during the first steps of the host oyster's immunological reaction, so far the fate of the graft and its exact role in the process of pearl formation could not be determined via classical histological methods. Genetic structure at different spatial scales in the pearl oyster (Pinctada margaritifera cumingii) in French Polynesian lagoons: beware of sampling strategy and genetic patchiness ArchiMer

In order to study further the genetic structure of the pearl oyster Pinctada margaritifera in French Polynesia with a special consideration for the sampling scale, we analyzed or re-analyzed sets of data based on nuclear DNA markers obtained at different spatial scales. At a large scale (several 1,000 km), the remote Marquesas Islands were confirmed to be significantly differentiated from Tuamotu-Gambier and Society archipelagos, with a marked difference however for the two main islands that are different from each other. Crossing phenotype heritability and candidate gene expression in grafted black-lipped pearl oyster Pinctada margaritifera, an animal chimera ArchiMer

Grafting mantle tissue of a donor pearl oyster into the gonad of a recipient oyster results in the formation of a chimera, the pearl sac. The phenotypic variations of this chimera are hypothesized to be the result of interactions between the donor and recipient genomes. In this study, the heritability of phenotypic variation and its association with gene expression were investigated for the first time during P. margaritifera pearl production. Particulate matter in Takapoto lagoon waters: potential food for cultivated pearl oysters ArchiMer

Results of research concerning the food of the pearl oyster Pinctada mai margaritifera are presented, by taking Takapoto atoll (Tuamotu archipelago, French Polynesia) as a study site. Monthly sampling of several parameters representative of bivalve environment and food (water temperature, salinity, mineral and organic matter, proteins, lipids and carbohydrates, chlorophyll a and phaeopigments) confirm and increase knowledge of the Takapoto lagoon hydrobiology. The variability of these parameters was analysed. Takapoto lagoon showed a remarkable spatial stability. Temporal variations showed a low seasonal trend. A study of pearl oysters Pinctada margaritifera growth and mortality was done on a monthly basis between June 1990 and September 1991 in Takapoto atoll (Tuamotu archipelago). Meanwhile, sampling parameters representative of bivalves food showed a remarquable spatial stability whithin the lagoon. The amplitude of temporal variability varied in the order of some days. Carrying capacity appears low : energetic content of particulate material in waters is about 2 joules/liter, ten times lower than in temperate areas. On the opposite, seasonal effect is reduced and particulate food is always available. This study was designed to measure responses of four-year-old black-lip pearl oysters (Pinctada margaritifera) to different temperatures and food concentrations and to identify the energy requirements of each sex. Oysters were fed a monospecific microalgal diet of Isochrysis affinis galbana (T-iso). Measurements of oxygen consumption and ingestion rates were carried out at 24 degrees C and 27 degrees C and at two algal (T-iso) concentrations: 5000 and 30 000 cell ml(-1). The shell biomineralisation with special reference to the nacreous region is observed during the development of the Polynesian pearl oyster. Ultrastructural changes were studied and timed for the first time from planktonic larval shells to two-year-old adult shells. During the first two weeks following fertilization, the prodissoconch-I shell structure is undifferentiated and uniformly granular. The prodissoconch-II stage which develops during the next two weeks acquires a columnar organization. A histological examination of grafting success in pearl oyster Pinctada margaritifera in French Polynesia ArchiMer

Pearl oyster grafting is a complex surgical operation that should lead to pearl formation after approximately eighteen months. Although this technique has been used for many years in French Polynesia, the grafting process is still not standardised. Cultured pearls are human creations formed by inserting a nucleus and a small piece of mantle tissue into a living shelled mollusc, usually a pearl oyster. Although many pearl observations intuitively suggest a possible rotation of the nucleated pearl inside the oyster, no experimental demonstration of such a movement has ever been done. This can be explained by the difficulty of observation of such a phenomenon in the tissues of a living animal. Hatchery-scale trials using cryopreserved spermatozoa of black-lip pearl oyster, Pinctada margaritifera ArchiMer

Cryopreservation is a valuable tool for genetic improvement programs. Several bivalve mollusc species have already been the subject of such programs and the Tahitian black pearl oyster industry is now planning the development of selective breeding for desirable traits in Pinctada margaritifera. The ability to cryopreserve spermatozoa would, therefore, offer significant benefits to the cultured black pearl industry. Spermatozoa were cryopreserved with CPA 0.7 M trehalose in 0.8 M Me2SO and a two-step freezing process was used: straws were first maintained in nitrogen vapour for 10 minutes, then directly plunged into liquid nitrogen and stored for one week before use. Phenotype plasticity, local adaptation, and biofouling influence on growth of the pearl oyster Pinctada margaritifera: A common garden approach ArchiMer

The purpose of our study is to investigate (1) the influence of phenotype plasticity and local adaptation on pearl-oyster physiology by testing the persistence of growth differentiation of two pearl oyster populations (Arutua and Mangareva) in common garden experiment; (2) to quantify the influence of biofouling development on the growth of each pearl oyster population.
